Ham’s Nutrient Mixture F-12 and Dulbecco’s Modified Eagle Medium are combined in DMEM/F-12 at a 1:1 ratio. L-glutamine is added to this DMEM/F-12 mix.
DMEM contains a buffer to maintain pH levels and usually comes with phenol red as a pH indicator. Cells cultivated in DMEM, or any other medium containing the bicarbonate buffer system, must be kept in a controlled CO2 environment of 5-10%. When phenol red is added, you can monitor pH changes from 6.2 (yellow) to 8.2 (red).

DMEM promotes the growth of several mammalian cell types. Along with cell lines including HeLa, HEK 293, Cos-7, and PC-12, it may be used to produce primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells. The original formulation was used to cultivate embryonic mouse cells and included 1.0 g/L of glucose. It has undergone several modifications since then. Additionally, for cells with higher energy requirements, the glucose levels have been increased to 4.5 g/L.
