As a foundational media, DMEM High Glucose without L-Glutamine (Dulbecco’s Modified Eagle Medium) is frequently used in cell culture. It offers fundamental nutrients for the development of many mammalian cell lines, especially when FBS is added.A number of primary cell types, including fibroblasts, neurons, HUVECs, smooth muscle cells, and even cell lines, including HEK-293, HeLa, Cos-7, and PC-12, have been effectively grown in DMEM in the past. DMEM is available in a number of formulations, each with a unique component makeup. In DMEM’s original composition, there are 1.0 g/L. This item, DMEM High Glucose, does not include L-glutamine and has an enhanced glucose content of 4.5 g/L.
Based on Eagle’s Minimal Essential Medium (EMEM), Dulbecco’s Modified Eagle Medium offers a four-fold increased content of vitamins and amino acids. Supplementing DMEM with FBS, usually at a dosage of 10%, is a relatively popular technique. This is frequently required because the medium lacks the proteins, lipids, and growth hormones required for cell development. The buffer in DMEM helps to keep pH levels stable, and phenol red is typically included as a pH indicator. Cells grown in DMEM or any other media with a bicarbonate buffer system need to be maintained in a regulated CO2 atmosphere of between 5 and 10% carbon dioxide. You can observe how the pH changes from 6.2 (yellow) to 8.2 when phenol red is introduced (red).
